Overview

DX308 is a potent, selective dual inhibitor of cytochrome P450 26A1 and 26B1 (CYP26A1/B1), enzymes responsible for the clearance of all-trans-retinoic acid (atRA). By inhibiting these enzymes, DX308 increases endogenous atRA concentrations in the skin without directly interacting with retinoic acid receptors. This mechanism allows it to deliver the therapeutic benefits of retinoids while minimizing adverse effects typically associated with exogenous retinoid administration. Developed by DermaXon, DX308 is being investigated primarily as a topical treatment for keratinization disorders such as ichthyosis and other retinoid-responsive dermatological conditions. Preclinical studies have demonstrated favorable metabolic and safety profiles, minimal off-target activity, no genotoxicity or significant toxicity in animal models, and effective modulation of retinoid-responsive genes in both healthy and diseased skin models[1][2][3][4][5][6]. There are also indications that it may be explored for hearing loss and Parkinson's disease[4][7].
