DX416 (also known as DX-416) is a fully human monoclonal antibody discovered by Dyax Corp (now part of Takeda) using phage display technology. It is designed to potently and selectively inhibit activated Factor XII (Factor XIIa), a serine protease that plays a critical role in the initiation of the contact activation pathway of coagulation and the kallikrein-kinin system. By inhibiting Factor XIIa, DX416 aims to prevent the formation of bradykinin and the activation of the intrinsic coagulation cascade, making it a potential therapeutic candidate for hereditary angioedema (HAE) and the prevention of thrombosis without increasing bleeding risk. While it showed promise in preclinical models, development appears to have been deprioritized in favor of other assets like lanadelumab (DX-2930).
