DXC006 + immune checkpoint inhibitor is an investigational combination therapy being developed for the treatment of small cell lung cancer (SCLC). DXC006 is an antibody-drug conjugate (ADC) developed by Hangzhou DAC Biotechnology that targets Delta-like ligand 3 (DLL3), a protein frequently overexpressed in SCLC and other neuroendocrine tumors. The ADC comprises a DLL3-specific monoclonal antibody conjugated to a potent cytotoxic payload, typically a DNA topoisomerase I inhibitor (such as an exatecan derivative), via a cleavable linker. By combining DXC006 with an immune checkpoint inhibitor (ICI) targeting the PD-1 or PD-L1 pathway, the regimen aims to achieve synergistic anti-tumor effects through direct ADC-mediated cytotoxicity and the restoration of anti-tumor immunity. This combination is currently under evaluation in Phase Ib/II clinical trials (NCT06344104) to assess its safety, tolerability, and preliminary efficacy in patients with SCLC.
