DXC018 is a novel bispecific antibody-drug conjugate (ADC) developed by Hangzhou DAC Biotechnology (DAC Biotech) for the treatment of HER2-expressing solid tumors, including gastrointestinal malignancies. The drug features a bispecific antibody backbone that targets two distinct epitopes of the Human Epidermal Growth Factor Receptor 2 (HER2), specifically domains II and IV, with high affinity. This dual-epitope binding strategy is designed to enhance target specificity and promote efficient internalization. DXC018 is distinguished by its dual-payload delivery system, which incorporates both an antimetabolite inhibitor and a topoisomerase I inhibitor. This combination of payloads is intended to induce potent DNA damage and cytotoxic effects through multiple mechanisms of action. In preclinical studies, DXC018 has demonstrated superior or comparable antitumor activity in various human cancer xenograft models.
