Overview

DY131 (also known as GSK9089) is a synthetic small molecule that acts as a selective agonist for the orphan nuclear receptors estrogen-related receptor beta (ERRβ/ESRRB) and estrogen-related receptor gamma (ERRγ/ESRRG). Research indicates that DY131-mediated activation of the ERRβ2 isoform in glioblastoma cells disrupts actin cytoskeletal dynamics by enhancing the interaction between ERRβ2 and cortactin, thereby inhibiting tumor cell migration and vesicle trafficking. Additionally, DY131 has been shown to modulate the tumor microenvironment by shifting tumor-associated macrophage polarization from a pro-tumor M2 phenotype toward an anti-tumor M1 phenotype. It is widely utilized as a chemical probe in oncology and metabolic research.
