Dyclonine is a small molecule topical anesthetic primarily used for the temporary relief of pain and irritation in the mouth and throat. It is commonly found in over-the-counter lozenges (such as Sucrets) and topical solutions (such as DycloPro), where it acts to suppress the gag reflex prior to procedures or provide symptomatic relief from sore throats, canker sores, fever blisters, oral ulcers, stomatitis, and ano-genital lesions[1][3][4][7]. Its mechanism of action involves blocking both the initiation and conduction of nerve impulses by decreasing neuronal membrane permeability to sodium ions. This stabilizes the membrane reversibly and inhibits depolarization, resulting in failure of propagated action potentials—effectively producing local anesthesia[1]. The main molecular targets are sodium channel protein type 10 subunit alpha (SCN10A) and sodium channel protein type 5 subunit alpha[1]. Dyclonine is absorbed through mucous membranes with rapid onset when applied topically. It has also been used in veterinary medicine for similar purposes[6].
