Overview

Dyngo-4a is a synthetic small-molecule inhibitor of dynamin GTPases, developed as an optimized, less cytotoxic structural analog of dynasore with markedly improved potency against helical, liposome-stimulated dynamin assemblies. It inhibits dynamin 1 and dynamin 2 with submicromolar to low micromolar IC50 values and potently blocks clathrin-mediated endocytosis, transferrin uptake, synaptic vesicle endocytosis, and activity-dependent bulk endocytosis in diverse cellular systems. By interfering with dynamin-dependent membrane fission and also inserting into the plasma membrane to alter lipid packing, Dyngo-4a is widely used as a research tool to study endocytosis, neurotransmission, toxin and viral uptake, and signaling, and has shown preclinical activity in models of botulinum neurotoxin intoxication, rotavirus infection, neuroblastoma cell differentiation, and leukemia cell death.
