Overview

Dyno-4z2 is a novel adeno-associated virus (AAV) capsid engineered for gene therapy applications targeting the retina. It is optimized for delivery to the retina via intravitreal (IVT) injection and demonstrates exceptional efficiency in transducing bipolar cells within the inner nuclear layer (INL). In non-human primate models, Dyno-4z2 achieves over 40-fold improvement in broad retinal transduction compared to AAV2 at clinically relevant doses, and a 5-fold improvement in bipolar cell transduction versus reference externally engineered capsids. Its enhanced performance supports its use in optogenetic ocular gene therapies aimed at restoring vision for patients with retinal degeneration. The capsid is designed for easy administration through IVT injection, enabling broader patient access compared to more invasive subretinal approaches, and it is manufacturable at large scale using AAV2-based processes[1][2][3].
