Overview

Dyno-bn8 is a novel AI-engineered adeno-associated virus (AAV) capsid developed by Dyno Therapeutics for systemic intravenous delivery of gene therapies targeting skeletal and cardiac muscle. In non-human primate studies, it achieves high transduction efficiency—81% of skeletal muscle cells and 20% of cardiac cells—at a low dose of 5.2e12 vg/kg, representing a 25-fold dose reduction compared to existing muscle gene therapies while featuring substantial liver detargeting to enhance safety by minimizing off-target accumulation and potential toxicity. Its muscle tropism is mediated by interaction with a highly conserved receptor shared between non-human primates and humans, with in vitro validation in human muscle cell lines supporting clinical translatability; it is also compatible with scalable AAV9-based manufacturing processes.[1][3][6]
