DZ1 is a phosphorothioate-modified '10-23' DNAzyme designed to target the mRNA of the Epstein-Barr virus (EBV)-encoded latent membrane protein-1 (LMP1). Developed by researchers at Xiangya Hospital of Central South University, DZ1 functions as a catalytic DNA molecule that specifically binds to and cleaves LMP1 mRNA, thereby downregulating the expression of the LMP1 protein. LMP1 is a primary oncogene in nasopharyngeal carcinoma (NPC) that drives tumor progression by activating signaling pathways such as NF-κB, AP-1, and STAT3. By suppressing LMP1, DZ1 inhibits cell proliferation and metastasis, promotes apoptosis, and significantly enhances the radiosensitivity of NPC cells. It has been investigated in Phase I/II clinical trials as an intratumoral adjunct to radiotherapy for patients with nasopharyngeal carcinoma.
