Overview

DZ1462 is a novel, **gastrointestinal-restricted small molecule inhibitor** targeting the **sodium-dependent phosphate cotransporter type 2b (NPT2b)**, which mediates intestinal phosphate absorption. By inhibiting NPT2b, DZ1462 blocks dietary phosphate uptake through the intestinal wall without significant systemic absorption, thus reducing serum phosphate and hyperphosphatemia. In preclinical models of chronic kidney disease (CKD) and hyperphosphatemia, DZ1462 showed potent efficacy, outperforming established phosphate binders like sevelamer, and also demonstrated reductions in cardiovascular events and bone loss. DZ1462 may be particularly useful for CKD patients unresponsive to conventional phosphate binders.
