**DZNep + GL-V9** is a combination of two small molecule agents: DZNep (3-deazaneplanocin A), an inhibitor of S-adenosylhomocysteine hydrolase (SAH hydrolase) that disrupts the histone methylation cycle by downregulating EZH2 and reducing H3K27me3 levels, and GL-V9, a synthetic flavonoid derivative of wogonin with antitumor activity. The combination exhibits strong synergistic effects in preclinical models of acute myeloid leukemia (AML) and pancreatic ductal adenocarcinoma (PDAC), inducing mitotic catastrophe, gross DNA damage response, apoptosis, cell cycle arrest (e.g., G1-S boundary or reduced G2/M), reduced migration via E-cadherin upregulation, and depletion of cancer stem cell markers like CD133. In AML, it inhibits HELLS (a DNA helicase), extends survival in xenograft models (e.g., U937 CDX), and reduces spleen size/weight; no clinical development or specific developers identified.
