E-4010 is a newly synthesized, potent, and selective orally active phosphodiesterase 5 (PDE5) inhibitor. It has demonstrated efficacy in attenuating hypoxic pulmonary hypertension in rats by causing a long-lasting reduction in mean pulmonary arterial pressure without significant effects on systemic arterial pressure, cardiac output, or heart rate. Long-term treatment with E-4010 also reduced right ventricular hypertrophy and pulmonary arterial remodeling, likely by increasing cGMP levels in the pulmonary vascular smooth muscle.
