E-612 is a bi-sialidase antibody-enzyme fusion protein developed by Palleon Pharmaceuticals for the treatment of autoimmune diseases. It is engineered using the company's proprietary EAGLE (Enzyme-Antibody Glycan Ligand Editing) platform. The drug is designed to enzymatically remove terminal sialic acids from cell-surface glycans, which act as immune-modulating signals. By desialylating these glycans, E-612 aims to disrupt the 'glyco-immune checkpoint' and restore immune homeostasis in patients with autoimmune disorders. As of the latest reports, the program is in the lead optimization stage of preclinical development.
