E-EDV-Dox (also known as EGFR-EDV-Dox) is an investigational targeted nanocell-drug delivery system developed by EnGeneIC Pty Ltd. It consists of EnGeneIC Dream Vectors (EDVs), which are ~400 nm bacterially-derived minicells produced from Salmonella typhimurium. These minicells are loaded with the cytotoxic chemotherapy agent doxorubicin and are surface-functionalized with bispecific antibodies that target the epidermal growth factor receptor (EGFR), specifically utilizing the panitumumab (Vectibix) sequence. The system is designed to deliver high concentrations of doxorubicin directly into EGFR-overexpressing tumor cells via receptor-mediated endocytosis, thereby minimizing systemic toxicity. E-EDV-Dox is primarily being investigated for the treatment of recurrent glioblastoma multiforme (GBM), often in combination with EDV-GC (minicells carrying alpha-galactosyl ceramide) to simultaneously activate an anti-tumor immune response.
