E10030 + aflibercept + bevacizumab is a combination therapy consisting of three agents targeting angiogenesis, primarily for ophthalmic and oncologic indications. \n- **E10030** (also known as Fovista) is an anti-platelet-derived growth factor-B aptamer that inhibits PDGF-B, aiming to prevent pericyte recruitment and vessel stabilization in neovascularization. \n- **Aflibercept** (VEGF Trap-Eye; brand name Eylea) is a recombinant fusion protein acting as a decoy receptor for vascular endothelial growth factor-A (VEGF-A), VEGF-B, and placental growth factor (PlGF), thereby inhibiting angiogenesis. \n- **Bevacizumab** (brand name Avastin) is a humanized monoclonal antibody that binds to VEGF-A, preventing its interaction with receptors on the surface of endothelial cells and thus inhibiting new blood vessel formation.\n\nThis triple combination targets multiple pathways involved in abnormal blood vessel formation—PDGF signaling via E10030 and VEGF signaling via aflibercept and bevacizumab—with the goal of more effectively suppressing pathological angiogenesis than monotherapy or dual therapy alone. The primary indications under investigation have included neovascular age-related macular degeneration (wet AMD) and other diseases characterized by pathologic ocular or tumor angiogenesis[6][9][10].
