E10A + pembrolizumab (also referred to as E10A + PD1) is an investigational combination therapy developed by Guangzhou Double Bioproducts. It combines E10A, a replication-deficient recombinant adenovirus type-5 vector encoding the human endostatin gene, with pembrolizumab, a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor. E10A is administered intratumorally to deliver the endostatin gene directly into tumor cells, where it expresses endostatin to inhibit endothelial cell proliferation, disrupt tumor angiogenesis, and block blood supply. Pembrolizumab is administered intravenously to block the PD-1/PD-L1 pathway, restoring T-cell-mediated antitumor immune responses. This dual mechanism synergistically targets both tumor vasculature and immune evasion. The combination is currently in Phase II clinical development for the treatment of metastatic or unresectable recurrent head and neck squamous cell carcinoma (HNSCC).
