Overview

E2025 is a humanized monoclonal antibody developed by Eisai that specifically targets Ephrin type-A receptor 4 (EphA4), a member of the ephrin receptor subfamily predominantly expressed in the brain. EphA4 activation is implicated in neurodegeneration associated with diseases such as Alzheimer's disease. By binding directly to EphA4, E2025 acts as an antagonist, inhibiting ligand binding and enhancing cleavage of EphA4. Preclinical studies have shown that E2025 suppresses neurodegeneration and progression of tau pathology in models of Alzheimer's disease by modulating the EphA4 pathway. The drug is being developed primarily for Alzheimer's disease and may promote synaptic remodeling of glutamatergic neurons[1][3][7].
