E2511 is an investigational small molecule drug developed in-house by Eisai. It acts as a positive allosteric modulator (PAM) of the tropomyosin receptor kinase A (TrkA), which is the high-affinity receptor for nerve growth factor (NGF) located on neural cell membranes. By binding directly to TrkA, E2511 enhances specific trophic signaling of NGF without triggering unwanted NGF-associated adverse effects. This mechanism may promote recovery and synaptic remodeling of damaged cholinergic neurons and suppress cerebral atrophy caused by neurodegeneration, making it a potential therapeutic candidate for neurodegenerative diseases such as Alzheimer’s disease. Phase 1 clinical studies have demonstrated brain penetration and generally favorable safety in healthy volunteers[2][3][4][5][6][9].
