E40

E40 (Endoprotease 40) is a recombinant protein enzyme therapeutic developed by Nemysis Limited for the management of celiac disease and non-celiac gluten sensitivity. The active ingredient is a novel endopeptidase originally discovered in the soil actinomycete *Actinoallomurus* A8 and produced via recombinant technology. Administered orally, E40 functions by enzymatically degrading gluten and its immunogenic peptides, particularly the immunotoxic epitopes of α-gliadin, within the stomach. This degradation prevents the peptides from reaching the small intestine in an intact form, thereby reducing or eliminating the immune response triggered by inadvertent gluten consumption. E40 is designed to be resistant to gastric acidity and endogenous digestive enzymes like pepsin, maintaining its proteolytic activity in the gastrointestinal tract. It is being developed as a Class IIb Medical Device in Europe.
