Overview

E6 T cell receptor therapy is an investigational adoptive cell therapy in which a patient's own (autologous) peripheral blood lymphocytes are genetically engineered to express a high-affinity T-cell receptor (TCR) that specifically recognizes the human papillomavirus type 16 (HPV-16) E6 oncoprotein presented by HLA-A*02:01 molecules. The modified cells are expanded ex vivo and reinfused into the patient to target and kill HPV-16+ cancer cells. This approach is being developed primarily for the treatment of advanced or metastatic cancers associated with persistent HPV-16 infection, such as cervical carcinoma and other epithelial cancers. The mechanism of action involves direct recognition and cytotoxic elimination of tumor cells expressing the viral E6 antigen[2][5][7][8].
