E6005 (also known as RVT-501) is a novel, small-molecule phosphodiesterase 4 (PDE4) inhibitor developed for the topical treatment of atopic dermatitis. By selectively inhibiting the PDE4 enzyme, the drug increases intracellular levels of cyclic adenosine monophosphate (cAMP), which subsequently suppresses the production of various pro-inflammatory cytokines and mediators involved in skin inflammation and pruritus. E6005 is formulated as a topical ointment and has been investigated in concentrations ranging from 0.01% to 0.3%. Clinical studies in both adult and pediatric populations have demonstrated that the drug improves skin eruptions and itching with very low systemic exposure, suggesting a favorable safety profile for mild to moderate lesions. The compound was originally discovered by Eisai and has been further developed through partnerships with companies including Roivant Sciences (Dermavant) and Organon and Co.
