Overview

E7 TCR is an autologous cell therapy in which a patient's own T lymphocytes are genetically engineered to express a high-avidity T cell receptor (TCR) specific for the human papillomavirus type 16 (HPV16) E7 antigen presented by HLA-A*02:01. The modified cells are expanded and reinfused into the patient to target and kill HPV16-positive cancer cells. The mechanism of action involves recognition of the tumor-restricted viral oncoprotein E7, leading to targeted cytotoxicity against infected or transformed cells while sparing normal tissues. This approach is being developed primarily for metastatic or refractory cancers associated with HPV16 infection, including cervical cancer, head and neck squamous cell carcinoma, anal cancer, vulvar cancer, vaginal cancer, penile cancer, and other anogenital cancers[1][2][3][4][5]. Clinical trials have demonstrated tumor regression in some patients with manageable toxicity profiles[3][6].
