E7011 is a first-in-class neutralizing monoclonal antibody that specifically targets the Notch4 receptor. Developed by Eisai, it is designed to inhibit Notch4 signaling, which is implicated in breast tumor initiation, the maintenance of cancer stem cells, and the regulation of the tumor microenvironment (TME). By blocking Notch4, E7011 reprograms tumor-associated macrophages (TAMs)—specifically the immunosuppressive Stab1+ subpopulation—into a pro-inflammatory, tumor-destructive phenotype. This functional reprogramming, along with a decrease in myeloid-derived suppressor cells (MDSCs), enhances anti-tumor immunity and reduces tumor growth in various models, including mammary carcinoma.
