E7016 is an orally active poly(ADP-ribose) polymerase (PARP) inhibitor that enhances tumor cell radiosensitivity by inhibiting DNA repair. When combined with temozolomide, an oral alkylating agent used primarily to treat glioblastoma multiforme and anaplastic astrocytoma, the combination has shown greater than additive effects in preclinical models, particularly in enhancing the efficacy of radiotherapy for brain tumors. Temozolomide works by damaging DNA in cancer cells, preventing their replication and growth. The combination is being investigated as a potential therapy to improve outcomes for patients with glioblastoma multiforme and other advanced solid tumors[1][2][3][4][5][7].
