E7389-LF + nivolumab is an **investigational combination therapy** comprising E7389-LF, a liposomal formulation of eribulin (a microtubule dynamics inhibitor), and nivolumab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1). E7389-LF provides cytotoxicity via microtubule inhibition and additionally modulates the tumor microenvironment, including vascular remodeling and immune activation. Nivolumab blocks PD-1, releasing immune inhibition to promote tumor-specific cytotoxicity. This combination has demonstrated preliminary antitumor activity, especially in patients with advanced or pretreated **gastric and esophageal cancers**. The combination exploits synergistic potential through direct cytotoxic effects and enhanced immune response, as observed in clinical and preclinical studies. Developed by Eisai (E7389-LF) and Bristol Myers Squibb/Ono Pharmaceutical (nivolumab), the regimen is not yet approved and remains in clinical investigation[1][2][3][4][5].
