EA-2 is a preclinical-stage tolerogenic biologic developed by Endo Axiom, primarily aimed at the prevention of Type 1 diabetes onset. The therapeutic utilizes a quantum-dot-enabled delivery platform designed for oral administration. Its mechanism of action involves the targeted delivery of tolerogenic signals to hepatic antigen-presenting cells (APCs), which are central to the induction of systemic immune tolerance. By promoting an immune-tolerant environment in the liver, EA-2 is designed to prevent the autoimmune destruction of pancreatic beta cells. The program has demonstrated efficacy in NOD (non-obese diabetic) mouse models and is also being explored as a platform for other conditions, such as peanut allergy.
