EA5 is a humanized monoclonal antibody developed by Shanghai Lanyi Therapeutics for the treatment of paroxysmal nocturnal hemoglobinuria (PNH). It functions as a complement inhibitor by specifically targeting complement component 5 (C5). By binding to C5, EA5 prevents its enzymatic cleavage into the pro-inflammatory mediator C5a and the initiator of the terminal complement pathway, C5b. This blockade prevents the formation of the membrane attack complex (MAC), thereby protecting PNH-affected red blood cells from intravascular hemolysis. EA5 is currently being evaluated in Phase Ib clinical trials to determine its safety, pharmacokinetic, and pharmacodynamic profiles in adult patients with PNH.
