**EAGLE-Her2** is an investigational HER2-targeted **antibody-sialidase fusion biologic** being developed by **Palleon Pharmaceuticals**. It consists of **trastuzumab** fused to an engineered **human sialidase** enzyme and is part of Palleon's **Enzyme-Antibody Glyco-Ligand Editing** platform. The molecule is designed to bind **human epidermal growth factor receptor 2** on HER2-expressing tumor cells and locally remove cell-surface **sialic acids**, thereby disrupting the **Siglec sialoglycan glyco-immune checkpoint** and promoting anti-tumor immunity. In preclinical HER2-positive breast cancer models, it has been reported to induce tumor desialylation and enhance innate immune mechanisms such as macrophage phagocytosis and dendritic cell activation. The program appears to be a preclinical oncology asset rather than an approved medicine.
