EB-DT-CAR-NK is an allogeneic, dual-target chimeric antigen receptor natural killer (CAR-NK) cell therapy developed by Beijing Biotech for the treatment of advanced or metastatic breast cancer. The therapy is designed to be biomarker-guided, where patients are treated with a specific dual-target CAR-NK product tailored to their tumor's antigen profile. The primary targets include Human Epidermal Growth Factor Receptor 2 (HER2/ERBB2), Mucin 1 (MUC1), and Receptor Tyrosine Kinase Like Orphan Receptor 1 (ROR1), with Mesothelin (MSLN) also considered for certain triple-negative breast cancer (TNBC) cases. By utilizing NK cells rather than T cells, the therapy aims to provide an 'off-the-shelf' allogeneic option with a potentially lower risk of graft-versus-host disease (GvHD). The treatment protocol typically involves a short course of lymphodepleting chemotherapy with fludarabine and cyclophosphamide prior to intravenous infusion of the CAR-NK cells to optimize cell persistence and anti-tumor activity.
