Overview

Ebastine is a second-generation long-acting nonsedating antihistamine of the piperidine class that acts as a selective antagonist of peripheral histamine H1 receptors. It is primarily indicated for the treatment of allergic rhinitis (including seasonal and perennial forms) and chronic idiopathic urticaria. By blocking H1 receptors in peripheral tissues without significant penetration into the central nervous system or antimuscarinic effects, ebastine effectively reduces symptoms such as sneezing, runny nose, itchy eyes and hives with minimal risk of drowsiness or cognitive impairment. After oral administration it undergoes extensive first-pass metabolism by hepatic cytochrome P450 3A4 to its active metabolite carebastine. Ebastine has demonstrated efficacy comparable to other second-generation antihistamines in clinical trials and is generally well tolerated with a safety profile similar to placebo; it does not prolong QT interval at recommended doses[1][2][4][5][6][8].
