EBC-129 is an investigational antibody-drug conjugate (ADC) developed in Singapore, designed to selectively target cancer cells across a broad range of solid tumors. The drug consists of a monoclonal antibody that binds to a specific N-glycosylated epitope conserved on both CEACAM5 and CEACAM6, which are cell surface proteins overexpressed in many cancers but not in normal tissues. This unique targeting allows EBC-129 to address cancers such as lung, gastric, oesophageal, pancreatic, breast, ovarian, and biliary tract cancers—including those with limited treatment options. Upon binding to its target on the cancer cell surface, the ADC is internalized and releases its cytotoxic payload—monomethyl auristatin E (MMAE)—within the tumor cell. MMAE disrupts microtubule function leading to cell death. The specificity for glycosylated CEACAM5/6 enables high tumor selectivity while sparing normal cells. Preclinical data suggest synergy with PD-1 inhibitors. As of 2024–2025, EBC-129 is undergoing Phase 1 clinical trials[1][3][4][5][6][8].
