EBD-300 is a novel, mammalian-derived recombinant asparaginase engineered to lack glutaminase activity. Developed by Everest Medicines, it is designed to address the limitations of traditional bacterial-derived asparaginases (such as those from *E. coli* or *Erwinia*), which often cause significant clinical toxicities—including hepatotoxicity, pancreatitis, and coagulopathy—due to their inherent glutaminase activity and high immunogenicity. EBD-300 works by depleting systemic levels of L-asparagine, an amino acid that certain leukemic cells are unable to synthesize internally. This depletion induces metabolic stress and apoptosis in cancer cells. Preclinical data suggest that EBD-300 exhibits synergistic anti-leukemic activity when combined with the BCL-2 inhibitor venetoclax, particularly in models of Acute Myeloid Leukemia (AML) harboring chromosome 7 deletions.
