Overview

**EBOV-515** is a human monoclonal antibody (mAb) isolated from Ebola virus survivors, designed as a pan-ebolavirus therapeutic. It targets the base region of the ebolavirus glycoprotein (GP), exhibiting potent neutralizing activity against Ebola virus (EBOV), Bundibugyo ebolavirus (BDBV), and Sudan ebolavirus (SUDV) with IC50 values ranging from 400 to 5,000 ng/mL. EBOV-515 demonstrates high efficacy in preclinical models, providing complete protection against lethal EBOV challenge in mice at doses as low as 1 mg/kg (especially the Fc-modified LALA-PG variant) and contributing to synergistic protection in nonhuman primates when combined with rEBOV-442. Its mechanism involves strong binding to cleaved GP (GPCL), inhibition of GP processing, and resistance to viral escape, positioning it as a promising candidate for post-exposure prophylaxis and treatment of ebolavirus disease.[1][4]
