**EBOV-520** is a human monoclonal antibody isolated from survivors of ebolavirus disease that binds to a conserved epitope in the base region of the ebolavirus glycoprotein (GP). It potently neutralizes Ebola virus (EBOV), Bundibugyo ebolavirus (BDBV), and Sudan ebolavirus (SUDV) with IC50 values ranging from 400 to 5,000 ng/mL, primarily through direct virus neutralization by inhibiting cleaved GP (GP~CL~) engagement with the endosomal receptor NPC1. EBOV-520 demonstrates enhanced binding (80- to 250-fold) and neutralizing potency (200- to 800-fold) against GP~CL~ compared to intact GP, confers complete protection in mouse models of EBOV infection at 5 mg/kg post-exposure, and is a candidate for pan-ebolavirus therapeutic cocktails, showing synergy with glycan-cap-specific antibodies like EBOV-548.[1][2]
