Ebrasodebart is a human IgG4 monoclonal antibody that antagonizes the binding of insulin-like growth factor binding protein 3 (IGFBP3) to the cell death receptor transmembrane protein 219 (TMEM219), which is expressed on intestinal stem cells and conserved across species. By inhibiting this interaction, ebrasodebart blocks transmembrane protein 219 (TMEM219)-mediated activation of Caspase-8 and prevents excessive apoptosis of intestinal stem cells—a process implicated in diseases such as ulcerative colitis. Ebrasodebart is being developed as a first-in-class investigational therapy for inflammatory bowel diseases, particularly ulcerative colitis and type 1 diabetes. The drug was initially developed by Enthera Srl and has reached Phase 1 clinical trials in healthy volunteers to assess safety, tolerability, pharmacokinetics, and predicted therapeutic concentrations[1][2][6].
