Overview

EBV-specific TCR-T cell with cytokine auto-secreting element is an investigational autologous cell therapy in which a patient's own T cells are genetically engineered to express a specific T-cell receptor (TCR) that targets Epstein-Barr virus (EBV)-infected cells. These engineered cells may also be modified to secrete immune-stimulatory cytokines upon activation, enhancing their anti-tumor activity and recruitment of additional immune effectors. The primary mechanism involves recognition and killing of EBV-positive tumor or infected cells via the introduced antigen specificity of the engineered TCR. This approach is being developed for the treatment of recurrent or metastatic EBV-positive cancers such as head and neck squamous cell carcinoma and extranodal NK/T-cell lymphoma[6][7]. Developers have explored further enhancements such as co-expression of inducible interleukin-12 (IL-12) to improve efficacy by modulating the tumor microenvironment[4].
