This combination immunotherapy regimen, developed by Endocyte, is designed for the treatment of progressive metastatic renal cell carcinoma (mRCC). The therapy exploits the high expression of folate receptors on RCC cells using a targeted immunotherapy approach. It consists of a priming phase with EC90 (a KLH-FITC conjugate) and the saponin adjuvant GPI-0100 to induce high levels of anti-fluorescein (anti-FITC) antibodies. This is followed by the administration of EC17 (folate-FITC), which binds specifically to folate receptors on the tumor surface, effectively decorating the cancer cells with FITC haptens. The pre-induced anti-FITC antibodies then bind to the tumor cells, mediating their destruction through antibody-dependent cellular cytotoxicity (ADCC). Low-dose cytokines, including interleukin-2 (IL-2) and interferon-alpha (IFN-alpha), are co-administered to enhance the activation and recruitment of natural killer (NK) cells and other immune effectors to the tumor site.
