ecdCD40L is a recombinant protein representing the extracellular domain of CD40 ligand (CD40L), used as an immunostimulatory component in cancer vaccines. Most notably, it is part of the Ad-sig-hMUC-1/ecdCD40L vaccine—a therapeutic cancer vaccine that uses a recombinant adenoviral vector to deliver both the tumor-associated antigen MUC1 and ecdCD40L. The mechanism involves stimulating dendritic cells via CD40 signaling to enhance anti-tumor immune responses against MUC1-expressing epithelial cancers. Preclinical and early clinical studies have shown that this approach can activate endogenous dendritic cells, induce potent anti-MUC1 immunity, and modulate the cancer immunome[2][4][8]. The primary indication has been for advanced adenocarcinomas such as breast, ovarian, lung, colon, and prostate cancers.
