ECI-006 is an investigational mRNA-based cancer vaccine developed for the treatment of melanoma. It consists of a combination of mRNAs encoding five different melanoma tumor-associated antigens (TAAs) and utilizes the TriMix platform, which includes three mRNAs encoding constitutively activated toll-like receptor 4 (caTLR4), CD40 ligand (CD40L), and CD70. This design aims to optimally activate dendritic cells, thereby stimulating a robust T-cell-mediated immune response against melanoma cells. The vaccine is administered via intranodal injection and has been evaluated both as monotherapy after tumor resection and in combination with anti-PD1 therapy in patients with metastatic melanoma. Clinical trials have demonstrated measurable immune responses against tumor antigens, though development appears to have been discontinued at phase 1[1][2][3][7][8].
