Overview

EcNΔnlpIIHCL (ENHL) is an engineered microbial therapy derived from the probiotic strain *Escherichia coli* Nissle 1917 (EcN). The bacterium is genetically modified with a deletion of the *nlpI* gene to enhance the biogenesis of outer membrane vesicles (OMVs), which serve as delivery vehicles for therapeutic payloads. ENHL features a bifunctional surface display system utilizing Ice Nucleation Protein (INP)-HlpA for tumor-specific targeting and ClyA-EGFP for visualization and tracking. It is further engineered with an arabinose-inducible system to express lactate oxidase (LOx) within the tumor microenvironment (TME). Administered orally, the bacteria selectively colonize tumor tissues where the localized expression of LOx depletes lactate, thereby neutralizing acidic stress, inhibiting aberrant DNA repair, and promoting immune cell infiltration. This metabolic reprogramming strategy is primarily designed to overcome radioresistance in colorectal cancer and enhance the overall efficacy of radiotherapy.
