EcN-Mel is an engineered probiotic derived from genetically modified Nissle 1917 Escherichia coli that expresses tyrosinase genes. This modification enables the bacteria to produce and secrete melanin. The melanin acts as a nanozyme, effectively scavenging reactive oxygen species, particularly at colitis lesions. EcN-Mel also works by modulating the gut microbiota, enhancing beneficial bacteria like Lactobacillus while decreasing harmful Proteobacteria, promoting gut microbiota homeostasis. Furthermore, it contributes to cytokine regulation and intestinal barrier restoration, collectively alleviating intestinal mucosal damage. It is being developed for the oral treatment of Ulcerative Colitis.
