EcNc-RtSOD is an engineered live biotherapeutic product (LBP) that utilizes the probiotic bacterium *Escherichia coli* Nissle 1917 (EcN) as a delivery chassis. The strain is modified with cryptic plasmids to constitutively express a superoxide dismutase (RtSOD), an antioxidant enzyme. Developed for the treatment of ulcerative colitis (UC), EcNc-RtSOD is designed to colonize the intestinal tract and provide a continuous supply of SOD directly to the inflamed mucosa. This localized delivery helps neutralize reactive oxygen species (ROS), thereby reducing oxidative stress and downregulating pro-inflammatory cytokines such as interleukin-6 (IL-6) and interleukin-1β (IL-1β). Preclinical studies in mouse models of colitis have demonstrated that treatment with EcNc-RtSOD significantly reduces intestinal inflammation, lowers malondialdehyde (MDA) levels, and promotes mucosal healing.
