Overview

EcNPAQ is an engineered live biotherapeutic product (LBP) consisting of a probiotic consortium based on the *Escherichia coli* Nissle 1917 (EcN) chassis. It is designed for cancer immunotherapy, specifically targeting colorectal cancer by integrating multiple signals from the tumor microenvironment (TME). The consortium utilizes an orthogonal quorum-sensing system to respond to three characteristic TME parameters: low pH, hypoxia, and high lactate levels. Upon activation, it achieves the programmed release of two therapeutic payloads: a lactate depletion enzyme (LdhA) to improve the metabolic environment and a programmed death ligand 1 (PD-L1) nanobody for immune checkpoint inhibition. Preclinical studies in humanized mouse models have demonstrated that EcNPAQ can restructure intestinal microbial ecology, enhance the infiltration of CD8+ and CD4+ T cells, and significantly suppress tumor progression.
