ECP-1014 is an orally bioavailable, third-generation cyclooxygenase-2 (COX-2) inhibitor developed for oncology indications by Euclises Pharmaceuticals. It is designed to be a more potent and selective COX-2 inhibitor than earlier drugs in this class, with the goal of achieving greater and more consistent suppression of COX-2 activity in tumors that overexpress this enzyme. Elevated COX-2 activity has been linked to cancer progression, resistance to standard therapies, and immune suppression. ECP-1014 is being evaluated primarily for the treatment of solid tumor cancers, with clinical trials focused on safety, tolerability, pharmacokinetics, and establishing an optimal dose that achieves at least a 70% reduction in COX-2 activity—a threshold associated with improved survival outcomes in prior studies using older inhibitors. ECP‑1014 was designed by Dr. John Talley (inventor of celecoxib) as a next-generation agent specifically optimized for oncology use; it is currently being studied in Phase 1 clinical trials for patients with solid tumors.
