Drug intelligence / Profile preview
ecto-TMEM219 is a recombinant protein consisting of the extracellular domain of the transmembrane protein 219 (TMEM219) receptor. It acts as a decoy receptor that binds to insulin-like growth factor binding protein 3 (IGFBP3), preventing IGFBP3 from interacting with endogenous TMEM219. The IGFBP3/TMEM219 axis is a pro-apoptotic pathway that triggers Caspase-8-mediated cell death in pancreatic beta cells, pancreatic endocrine progenitors, and intestinal stem cells (ISCs). By blocking this pathway, ecto-TMEM219 protects these stem and progenitor cells from apoptosis, promoting cell survival, self-renewal, and tissue regeneration. It is being investigated for the treatment of type 1 diabetes (T1D), type 2 diabetes (T2D), diabetic enteropathy, and inflammatory bowel diseases (IBD) such as Crohn's disease and ulcerative colitis.
