Overview

Eculizumab-aeeb is a recombinant humanized monoclonal IgG2/4κ antibody that acts as a complement inhibitor. It is produced in Chinese Hamster Ovary (CHO) cells and consists of human constant regions from IgG2 and IgG4, with murine complementarity-determining regions grafted onto the human framework. Eculizumab-aeeb specifically binds to the complement protein C5 with high affinity, inhibiting its cleavage into C5a and C5b, thereby preventing formation of the terminal complement complex (C5b-9). This action blocks terminal complement-mediated intravascular hemolysis in paroxysmal nocturnal hemoglobinuria (PNH), inhibits complement-mediated thrombotic microangiopathy in atypical hemolytic uremic syndrome (aHUS), and reduces terminal complement complex deposition at the neuromuscular junction in generalized myasthenia gravis (gMG). Eculizumab-aeeb is an interchangeable biosimilar to Soliris, approved for intravenous infusion for PNH, aHUS, and gMG[5][7][8].
