Edatrexate is a synthetic antifolate and structural analog of methotrexate. It was developed as an antineoplastic agent for the treatment of various cancers. Edatrexate acts by inhibiting dihydrofolate reductase (DHFR), thereby blocking DNA synthesis and cell division in rapidly proliferating cells. It was investigated primarily for use in non-small cell lung cancer and other solid tumors but did not achieve widespread clinical adoption due to limited efficacy compared to existing therapies and notable toxicity.
