EDC34 is a 34-amino acid bioactive peptide derived from the C-terminal domain of Tissue Factor Pathway Inhibitor-2 (TFPI-2). It is being investigated as a multi-functional therapeutic agent for the treatment of severe Gram-negative sepsis, including infections caused by *Escherichia coli* and *Pseudomonas aeruginosa*. EDC34 exhibits a dual mechanism of action: it exerts direct bactericidal activity against Gram-negative bacteria and enhances the classical complement pathway, specifically boosting the formation of the antimicrobial peptide C3a. Simultaneously, it acts as an immunomodulator by inhibiting the bacteria-induced activation of the contact system, which helps prevent excessive coagulation and reduces associated organ damage, such as lung injury. Preclinical studies in mouse models have demonstrated that EDC34 significantly improves survival rates and reduces bacterial load, particularly when administered in combination with conventional antibiotics like ceftazidime.
