Overview

Edelinontrine (CRD-740) is an oral small molecule inhibitor of phosphodiesterase 9 (PDE9) developed by Cardurion Pharmaceuticals for the treatment of heart failure, including heart failure with reduced ejection fraction (HFrEF) and heart failure with preserved ejection fraction (HFpEF). PDE9 is an enzyme that specifically degrades cyclic guanosine monophosphate (cGMP) generated by the natriuretic peptide receptor (NPR) pathway. By inhibiting PDE9, edelinontrine aims to augment NPR-linked cGMP signaling, which provides cardioprotective effects such as vasodilation and reduced cardiac hypertrophy. Although the drug demonstrated proof-of-mechanism by significantly increasing plasma cGMP levels in the Phase 2a CARDINAL-HF trial, its development has reportedly been terminated.
